Synthesis of C-branched spermine tethered oligo-DNA and the thermal stability of the duplexes and triplexes

The first synthesis of the new C-branched spermine derivative 17, as well as its ability to
stabilise DNA duplexes and triplexes, are reported. The C-branched spermine block 17 was
converted into the corresponding O-(2-cyanoethyl)-(N, N-diisopropyl) phosphoramidite block
18 for incorporation at the 5′-end of DNA. It was also coupled to the 2′ of ara-U through a
phosphate bridge, leading to the partially protected 3′-hydroxy block 23, which was either
